Using microsatellite markers, we compared the genetic structure of populations of two carabid species, one described as a generalist (commonly found in forest and in open habitats) and the other known as a forest specialist. Both species were sampled in the same forest plots, which were separated from each other by either open or forested areas. At the local scale considered (13.6 km separating the most distant plots), genetic differentiation was substantial for both species studied, but populations of the forest specialist Carabus punctatoauratus appeared to be more spatially structured than those of C. nemoralis. Isolation by distance analyses showed that nonforested areas are partial barriers to gene flow for both species studied, although more clearly for the forest specialist. Between and within forests, dispersal capacity of the generalist C. nemoralis was shown to be higher than that of the specialist C. punctatoauratus.  相似文献

Acanthoscelides Schilsky is a large genus of neotropical bruchid beetles, in which most species show host plant specialization. Acanthoscelides obtectus and Acanthoscelides obvelatus are two sibling species specialized on Phaseolus beans, and are therefore considered pests. Up to now, the status of these two taxa has remained unclear, the few studies conducted having failed to elucidate whether these are two differentiated species or a single morphologically variable species. In addition, A. obvelatus has not been taken into account in the great majority of studies of bean bruchids. In this morphological and genetic study, we show that A. obtectus and A. obvelatus are two 'true' non-hybridizing species, which diverged about 22 Mya. Although the two species demonstrate only few morphological differences, we point out some diagnostic characters that enable their identification in the field. We also address a genetic method of differentiation of the two species, based on species-specific microsatellite loci. The strong morphological resemblance of these two species, despite their ancient divergence, may be the result of evolutionary stasis, which could be the consequence of stabilizing selection. Niche differentiation could enable the two species to coexist indefinitely.  相似文献

Cynipids are known to use various reproductive modes (arrhenotoky, thelytoky and strict cyclical parthenogenesis), but the mechanism remains unknown. We have studied the reproductive system of a rose gallwasp, Diplolepis spinosissimae, which was found to exhibit two different reproductive systems according to the population. In eight out of the ten populations studied, all along the Atlantic coast, D. spinosissimae is thelytokous. Males are extremely rare, and all females are homozygous at three microsatellite loci. ''Obligate homozygous parthenogenesis'' was found to be strictly associated with the presence of the endosymbiotic bacterium Wolbachia sp. In the two remaining populations, deprived of Wolbachia, D. spinosissimae reproduced by arrhenotoky as indicated by the larger frequency of males and heterozygosity of females. Allelic diversity, although not zero, was highly reduced in the coastal populations, as a consequence of thelytoky and gamete duplication. We hypothesize that mating of uninfected males with infected females during the first generations after an infection event could explain the small but significant amount of polymorphism observed in those populations. The high level of differentiation indicates a low gene flow, even between geographically close coastal populations.  相似文献

The fragmentation and destruction of natural habitats is one of the major causes of extinction of wild flora and fauna due to human activities. The area around the Mediterranean Basin is particularly affected: this region is one of the world’s richest in terms of biodiversity and is undergoing substantial modification due to extensive changes in land use. We investigated the demographic state (through a genetic survey) of an endangered grasshopper, the geographical distribution of which is restricted to arid open land or glades in south-eastern France. We compared two subspecies: one is severely threatened by the fragmentation of its specific and restricted habitat (Prionotropis hystrix rhodanica in the ‘Crau’ steppe) whereas the other, P.␣h.␣azami, occurs more widely in glades or clearings in the evergreen Mediterranean forest. Genetic diversity at eight microsatellite loci was highly structured, indicating substantial isolation of populations. Within-population diversity was high at all but one site. Genetic drift was the major force involved in this genetic structure, with very little gene flow at the regional scale of observation, consistent with both the limited dispersal of this flightless species and the patchy configuration of its habitat. No significant differences in the extent of genetic diversity or population dynamics were observed between the two subspecies. The observed genetic pattern calls into question the validity of the subspecies status, and shows no strong impact of recent anthropogenic modification of the Crau steppe. The evolution of the Mediterranean xeric and open habitats under both natural and anthropogenic processes is discussed.  相似文献

Chalcidoidea (Hymenoptera) are extremely diverse with more than 23,000 species described and over 500,000 species estimated to exist. This is the first comprehensive phylogenetic analysis of the superfamily based on a molecular analysis of 18S and 28S ribosomal gene regions for 19 families, 72 subfamilies, 343 genera and 649 species. The 56 outgroups are comprised of Ceraphronoidea and most proctotrupomorph families, including Mymarommatidae. Data alignment and the impact of ambiguous regions are explored using a secondary structure analysis and automated (MAFFT) alignments of the core and pairing regions and regions of ambiguous alignment. Both likelihood and parsimony approaches are used to analyze the data. Overall there is no impact of alignment method, and few but substantial differences between likelihood and parsimony approaches. Monophyly of Chalcidoidea and a sister group relationship between Mymaridae and the remaining Chalcidoidea is strongly supported in all analyses. Either Mymarommatoidea or Diaprioidea are the sister group of Chalcidoidea depending on the analysis. Likelihood analyses place Rotoitidae as the sister group of the remaining Chalcidoidea after Mymaridae, whereas parsimony nests them within Chalcidoidea. Some traditional family groups are supported as monophyletic (Agaonidae, Eucharitidae, Encyrtidae, Eulophidae, Leucospidae, Mymaridae, Ormyridae, Signiphoridae, Tanaostigmatidae and Trichogrammatidae). Several other families are paraphyletic (Perilampidae) or polyphyletic (Aphelinidae, Chalcididae, Eupelmidae, Eurytomidae, Pteromalidae, Tetracampidae and Torymidae). Evolutionary scenarios discussed for Chalcidoidea include the evolution of phytophagy, egg parasitism, sternorrhynchan parasitism, hypermetamorphic development and heteronomy.  相似文献

The species of Pseudidarnes are revised, and six species are described: P. acaudus Farache & Rasplus, sp. n.; P. astridae Farache & Rasplus, sp. n.; P. badiogeminus Farache & Rasplus, sp. n.; P. cooki Farache & Rasplus, sp. n.; P. kjellbergi Farache & Rasplus, sp. n.; P. laevis Farache & Rasplus, sp. n. Pseudidarnes minerva Girault, 1927 and P. flavicollis Bouček, 1988 are redescribed. A key to the species is provided as well as illustrations for all females and all known males (except the wingless male of P. minerva). We also provided further discussion on ecology, morphological patterns, and host taxonomy. We isolated and characterized nine and five polymorphic microsatellite loci in the respective ground beetles Carabus punctatoauratus and C. solieri. We tested cross‐species amplification of all these loci plus six isolated in C. solieri and for which primers sequences were soon published. From these combined analyses, we obtained 14 and 17 polymorphic markers, respectively, for C. punctatoauratus and C. solieri.  相似文献

Microsatellite loci were isolated from the coffee berry borer, Hypothenemus hampei, which is regarded as the major pest in coffee cultures. Seven polymorphic loci were obtained from an enriched genomic library. A low to moderate genetic diversity was observed per locus, with an observed number of alleles ranging from two to five in the 39 unrelated individuals sampled from Ethiopia. A clear deficit of heterozygotes within the population (mean heterozygosities, HO = 0.10/HE = 0.50) and an extreme inbreeding (FIS, 0.70–1.00) were demonstrated. Cross‐species amplifications showed that some of the markers could be useful in two closely related Hypothenemus species.  相似文献

Populations on islands often exhibit lower levels of genetic variation and ecomorphological divergence compared to their mainland relatives. While phenotypic differentiation in characters, such as size or shape among insular organisms, has been well studied, insular differentiation in quantitative reproductive traits involved in chemical communication has received very little attention to date. Here, we investigated the impact of insularity on two syntopic bumblebee species pairs: one including species that are phylogenetically related (Bombus terrestris and B. lucorum), and the other including species that interact ecologically (B. terrestris and its specific nest inquiline B. vestalis). For each bumblebee species, we characterized the patterns of variation and differentiation of insular (Corsican) vs. mainland (European) populations (i) with four genes (nuclear and mitochondrial, 3781 bp) and (ii) in the chemical composition of male marking secretions (MMS), a key trait for mate attraction in bumblebees, by gas chromatography-mass spectrometry (GC-MS). Our results provide evidence for genetic differentiation in Corsican bumblebees and show that, contrary to theoretical expectations, island populations of bumblebees exhibit levels of genetic variation similar to the mainland populations. Likewise, our comparative chemical analyses of MMS indicate that Corsican populations of bumblebees are significantly differentiated from the mainland yet they hold comparative levels of within-population MMS variability compared to the mainland. Therefore, insularity has led Corsican populations to diverge both genetically and chemically from their mainland relatives, presumably through genetic drift, but without a decrease of genetic diversity in island populations. We hypothesize that MMS divergence in Corsican bumblebees was driven by a persistent lack of gene flow with mainland populations and reinforced by the preference of Corsican females for sympatric (Corsican) MMS. The impoverished Corsican bumblebee fauna has not led to relaxation of stabilizing selection on MMS but to consistent differentiation chemical reproductive traits on the island.  相似文献
